This review studies the structure, activation pathways, and specific cellular location of P2X7R in order to analyze its biological relevance. It points to a possible allosteric modulation of this receptor by endogenous mediators, which would turn the tiny regulated ATP inputs into an activated functional P2X7R signaling, supporting the relevance of this receptor in a non-damaged tissue.
